Neurodegenerative diseases: New findings on protein misfolding

ScienceDaily (Sep. 18, 2012) ? Misfolded proteins can cause various neurodegenerative diseases such as spinocerebellar ataxias (SCAs) or Huntington's disease, which are characterized by a progressive loss of neurons in the brain. Researchers of the Max Delbr?ck Center for Molecular Medicine (MDC) Berlin-Buch, Germany, together with their colleagues of the Universit? Paris Diderot, Paris, France, have now identified 21 proteins that specifically bind to a protein called ataxin-1. Twelve of these proteins enhance the misfolding of ataxin-1 and thus promote the formation of harmful protein aggregate structures, whereas nine of them prevent the misfolding.

Proteins only function properly when the chains of amino acids, from which they are built, fold correctly. Misfolded proteins can be toxic for the cells and assemble into insoluble aggregates together with other proteins. Ataxin-1, the protein that the researchers have now investigated, is very prone to misfolding due to inherited gene defects that cause neurodegenerative diseases. The reason for this is that the amino acid glutamine is repeated in the amino acid chain of ataxin-1 very often -- the more glutamine, the more toxic the protein. Approximately 40 repeats of glutamine are considered to be toxic for the cells.

Now, Dr. Spyros Petrakis, Dr. Miguel Andrade, Professor Erich Wanker and colleagues have identified 21 proteins that mainly interact with ataxin-1 and influence its folding or misfolding. Twelve of these proteins enhance the toxicity of ataxin-1 for the nerve cells, whereas nine of the identified proteins reduce its toxicity.

Furthermore, the researchers detected a common feature in the structure of those proteins that enhances toxicity and aggregation. It is a special structure scientists call "coiled-coil-domain" because it resembles a double twisted spiral or helix. Apparently this structure promotes aggregation, because proteins that interact with ataxin-1 and have this domain enhance the toxic effect of mutated ataxin-1. As the researchers said, this structure could be a potential target for therapy: "A careful analysis of the molecular details could help to discover drugs that suppress toxic processes."

The potential for you or me to become the next victim of cybercrime is something that we both need to think about. I?ve been a victim a few times and I can tell you that it is frightening to be on the short end of the cybercriminal stick.

Late one afternoon (in early 2011) my bank balance appeared to be dropping fast, and the only connection I could see was that it involved Megaupload Limited and?PayPal. I contacted both PayPal and the 800 number to my bank, but I honestly did not feel as though they were doing enough to resolve my situation. The theft was happening in real time! So I did the next best thing and jumped on Twitter and tweeted?that an account using ?Megaupload Limited?was draining my bank account via?paypal! (At that time my main bank account was still back east in New Hampshire).

I use desktops with Vista and Windows 7, an iMac, a laptop (Win 7), a Linux server, an iPhone and an Android. I find myself layering different operating systems with whatever flavor works for me. On Windows and iMac I generally use Cocoon in varying capacities and also use Cocoon on my iPhone as my primary browser of choice. On my Linux machine I am mainly inside my terminal and rarely use a browser. If I am on public Wi-Fi ? Cocoon is my top choice for browsing the Internet.

I also find times that I use other services such as TorProject, Abine, and Hotspot Shield. It all depends upon the nature of what I need to do online when I am away from home or traveling. I am a very strong advocate of Internet security and privacy and appreciate having a multitude of online tools to choose from!

When you open the box it comes in 7 sachets. ?One for each day. ?Each sachet comes with powder that you can put in your food or drink. ?It's white and looks like salt or sugar.

What is it?

No taste, but smells like talc or eyeshadow

Mostly yogurt or in coffee or tea

What are the cons?

Can't technically eat it with everything. ?I tried clear skin with most foods and the powder clumps together and creates lumps that doesn't look appetizing to eat and it also gets stuck in my teeth. ?If your going to eat it with food makes sure the food has a lot of texture to it. ?Ice cream was a big no no. ?The powder just clumped together with cold water and rice. ?Salad and yogurt was manageable. ?I would only suggest eating clear skin with hot foods, soups, or drinking it with hot coffee or tea.

?What are the results?

Here's a macroshot of my left cheek. ?The bigger acne scars have significantly reduced after a month, however my pores are still really big.

Hundreds of business people from across the South West attended the launch of the week-long?British Business Pavilion?Olympic legacy event at Weymouth College yesterday (Monday 6 August).

Legendary voice of the Saints Jim Henderson joins Fox 8 sports

Fox 8 is proud to announce the legendary "voice of the Saints", Jim Henderson, is the newest member of our sports team providing the station with his unique and in-depth analysis of the Saints.?

Henderson's?duties will include providing?his popular commentaries?every week during the Saints season. You will also be able to get Jim's thoughts on our Fox 8 morning show the day after every Saints game as well as a weekly web chat, a?preview of each Saints game?and?he'll be a regular guest on?our Fox 8 Live Tailgate shows and?Final Play on Sunday nights. Henderson?is very happy?in?semi-retirement so you won't see him on the?anchor desk.?

Jim?becomes the third member of?the Fox 8 sports team with network experience and says he's thrilled to join Fred Hickman, Jennifer Hale and Sean Fazende to provide sports fans with the best local sports coverage imaginable.

"I'm excited about the new challenge," said Henderson. "I'm just going to be a part of the veteran bench strength that Fox 8 already has in place and I'm just happy to be joining Fred, Jen and Sean.?I leave a lot of good friends?behind but I get reunited with a lot of good friends as well. There are an awful lot of people at Channel 8 that I know and have respected and known for a number of years."

Fox 8 President and General Manager Joe Cook is honored to have Henderson join his decorated sports staff.

News Director Mikel Schaefer has a long history with Henderson having worked with the veteran sportscaster for nearly 25 years, 12 of those as one of Jim's producers.

"I couldn't be more proud or humbled to have Jim join our sports team," said Schaefer. "It's an honor to have someone as credible, talented and beloved as Jim on Fox 8. Who doesn't want to know what the?voice of the Saints thinks about the team after a game. His weekly commentaries are legendary and fans will continue to get them on Fox 8."

?Henderson role as the Saints play by play announcer with WWL-AM will be unchanged and as far as working for Tom Benson's television station, Jim says that's not a problem for him.

?"If people say can you still be objective? Well in my role as Saints play by play man, he (Tom Benson), has to pass judgment on me every single year as to whether I would be retained or not. He had the right of refusal on the Saints broadcast team. Not only did he allow me to stay on since 1986 but I never got one word of advice, one word of a threat or any heavy handiness in any way and there were times he certainly could have."?

Since he arrived in New Orleans in 1978, Jim established himself as the preeminent sports voice in the Gulf South. But while Henderson's love affair with New Orleans sports fans?began more than three decades ago, it was an inauspicious start. When Henderson was set to replace then?sports legend "Hap" Glaudi,?sports fans picketed outside of the station. But it didn't take long?for Henderson to win over his detractors. With a smooth as silk broadcasting style,?uncharacteristic wit?and an unmatched ability to craft sports prose, Jim became a local legend.?

Never has his talent been more on display than his three decades calling?Saints games on the radio. He's given sports fans a great reason to "turn up the sound on your?radio." Henderson's radio calls are synonymous with Saints football and with the exception of a year?calling NFL network games,?his trusted perspective has been a part of Saints football for three decades.?

Henderson's ability to?describe the action is unparalleled and like every great announcer he has left Saints fans with some of the?most memorable lines ever. His most famous coming when the Saints?clinched a Super Bowl berth after?winning the NFC Championship game in 2009, "Pigs have flown! Hell has frozen over! Olympics 2012: Among the top gold winners in London ... Kazakhstan?

One of the world's least populated countries at around 15 million, Kazakhstan is No. 8 in gold medals at the London Olympic Games.

If you've been checking out the gold medal count recently, chances are you've noticed the sky blue flag with the yellow sun and bird hovering at the No. 8 slot.?That would belong to Kazakhstan, the ninth largest country in the world, and one of the least populated countries at around 15 million people. This year has been its best-ever Olympic Games.

With six gold medals and a bronze under its belt and five days left in the 2012 Olympics, Kazakhstan is placed eighth, ahead of Germany and Australia for gold medals. In terms of gold medals per capita, Kazakhstan ranks fifth worldwide. Not bad for a country that only began to compete in the Olympics in 1994, two years after it became independent.

No doubt decades of Soviet rule influenced not only architecture, education, and government in Kazakhstan, but also its effective approach to sports training.

So what, exactly, is Kazakhstan medaling in?

The horsemeat advantage

Ilya Ilin took the gold and broke a new world record for the men's 94 kilogram weightlifting class when he hefted 418 kilograms, 12 kilos more than he lifted in Beijing when he won gold for total lifted. The 24-year-old two-time Olympic champion attributes his success to kazy ? horsemeat sausage.?

Svetlana Podobedova, who left her home country, Russia, after she was cut from the national weightlifting team for doping in 2007, narrowly beat Russia's weightlifter in the 75 kilogram class.

Maiya Maneza won the women's 63 kilogram weightlifting class. At age 26, she set?a new Olympic record of 245 kg?when she added a 135 kg clean and jerk to her 110 kg snatch.?(She took gold medals in the 2010 Asian Games, the 2010 World Weightlifting Championships, and the 2009 World Weightlifting Championships).

Teenager Zulfiya Chinshanlo won the women's 53 kilogram division with a new world record, lifting 226 kg. That was?Kazhakstan's second gold of the 2012 Olympic Games.?

On the sidelines, a small birther controversy has erupted. There are mixed reports on where exactly Maneza and Chinshanlo were born. Some claim that they were born in China or Kyrgyzstan. The Central Asian nation has responded pretty quickly and adamantly to reports that Maneza and Chinshanlo were born and raised in China.

President Nursultan Nazarbayev rewarded the pair with $250,000 each and hailed their wins as "proud glories" from "outstanding daughters of our nation."

Running and cycling, too

The Central Asian nation has topped the charts in other sports too.?

Olga Rypakova picked up a track-and-field gold for Kazakhstan in the women's triple jump. It's a unique event that combines a hop, a step, and a flying leap. The 27-year-old's 14.98-meter effort was enough to surpass the favorite, Olha Saladuha of Ukraine.

And 38-year-old Alexandre Vinokourov cycled past Colombia's Rigoberto Uran in the final stages of the men's road race to take the gold, leaving Norway's Alexander Kristoff finishing third on July 28. Kazakhstan and Mr. Vinokourov are no strangers to cycling: Among the athlete's accomplishments are four stage wins of the Tour de France,?two bronze medals at the World Championships, a silver at the 2000 Summer Olympics,?and the fact that he has been a known challenger to Lance Armstrong in the past.?

Happy with his time of 5:45:57, he then announced he was considering retiring.?

But Kazakhstan won't likely be retiring from racking up the medals?any time soon.?Watch the upcoming boxing and wrestling matches for more of the shiny stuff, as Kazakhstan bests its 2008 Beijing performance. (Already,?Danyal Gajiyev has taken bronze?in the?men's 84kg Greco-Roman wrestling category.)

Kazakhstan came home from Beijing with two golds (boxing and weightlifting), four silvers (judo, weightlifting, and wrestling), and seven bronze medals (boxing, judo, taekwondo, weightlifting, and wrestling) to finish 29th overall.
